It enrolled 3,127 adults with obesity or overweight, and at 72 weeks the highest dose (36 mg) produced an average weight loss of 12.4% (about 27.3 lbs). Among participants on that dose, 59.6% lost at least 10% of their body weight and 39.6% lost at least 15%. The lead investigator framed it as strong enough to work as a first-line option in primary care, precisely because it is a pill.
The program runs deep. ATTAIN-2 tested orforglipron in adults with obesity and type 2 diabetes and delivered significant weight loss alongside meaningful A1C reductions and improvements in cardiometabolic risk factors. The ACHIEVE trials confirmed strong blood-sugar control in type 2 diabetes, and ATTAIN-MAINTAIN showed it could hold weight off after people transitioned from injectable therapy. Across the board, no liver safety signal was observed, and side effects were the mild-to-moderate gastrointestinal ones typical of the class.
