CJC-1295 and Ipamorelin: Growth Hormone Secretagogues

CJC-1295 and Ipamorelin are popular as a pair because they attack the same goal from two different directions. Both prompt the body to release its own growth hormone, but they pull separate levers in the pituitary, and when used together they produce a stronger, more natural growth hormone pulse than either achieves alone. For research into recovery, body composition, and the growth hormone axis, it is one of the most studied combinations in peptide science.

What they are

CJC-1295 is an analog of growth hormone-releasing hormone (GHRH). It is built from the first 29 amino acids of natural GHRH, the shortest sequence that still works, with modifications that extend its life in the body. The version with a Drug Affinity Complex (DAC) binds to albumin in the blood, stretching its half-life to roughly six to eight days for sustained signalling.

Ipamorelin is a different kind of compound: a pentapeptide that belongs to the growth hormone-releasing peptide (GHRP) class. It is a selective agonist of the ghrelin receptor (GHS-R1a), the same receptor the hunger hormone uses to trigger growth hormone release. What sets Ipamorelin apart from older GHRPs is its cleanliness. It releases growth hormone without the spikes in cortisol and prolactin that earlier compounds caused, and without driving appetite at research doses.

What the research shows

The case for using these two together rests on a well-documented synergy. Growth hormone release is governed by two separate systems in the pituitary: the GHRH pathway and the ghrelin/GHRP pathway. Research has shown that stimulating both at once produces a growth hormone response far greater than the sum of the two stimulated separately, an effect often described as GHRH+GHRP synergy.

On CJC-1295 specifically, a clinical study by Walker and colleagues (2006, Journal of Clinical Endocrinology and Metabolism) found that CJC-1295 with DAC produced sustained, dose-dependent increases in growth hormone and IGF-1 lasting several days from a single injection, with IGF-1 rising roughly 1.5 to 3 times above baseline in healthy adults. Pairing that steady GHRH signal with Ipamorelin’s sharp, selective pulse is what makes the combination so effective at mimicking the body’s natural growth hormone rhythm.

How it works

CJC-1295 binds the GHRH receptor on pituitary somatotrophs, raising cAMP and driving the synthesis and release of growth hormone over a long window. Ipamorelin binds the separate GHS-R1a (ghrelin) receptor, triggering a fast, clean pulse of growth hormone through a calcium-dependent pathway. Because the two receptors feed into growth hormone release through different mechanisms, hitting both at once amplifies the output and produces a pulse pattern closer to how a healthy body naturally releases growth hormone. Both ultimately raise IGF-1, the downstream driver of growth hormone’s effects on recovery and body composition.
