CJC-1295 is an analog of growth hormone-releasing hormone (GHRH). It binds the GHRH receptor on the pituitary, the same receptor the body’s natural GHRH uses, and tells the gland to produce and release growth hormone. In its DAC form, it binds to albumin in the blood, giving it a long half-life of several days for a steady, sustained signal.

Ipamorelin is a growth hormone-releasing peptide (GHRP). It works through a completely different receptor, the ghrelin receptor (GHS-R1a), to trigger a sharp, clean pulse of growth hormone. What sets it apart from older GHRPs is its selectivity: it releases growth hormone without the unwanted spikes in cortisol and prolactin that earlier compounds caused.

CJC-1295’s signature is sustained elevation. In a clinical study by Walker and colleagues (2006, Journal of Clinical Endocrinology and Metabolism), CJC-1295 with DAC produced dose-dependent increases in growth hormone and IGF-1 that lasted several days from a single injection, with IGF-1 rising roughly 1.5 to 3 times above baseline.

Ipamorelin’s signature is a precise, well-tolerated pulse, valued specifically because it leaves cortisol and prolactin largely untouched and does not drive appetite at research doses.

The most important finding is what happens when they are combined: stimulating both the GHRH and the ghrelin pathways at once produces a growth hormone response greater than the sum of each alone, a well-documented effect known as GHRH+GHRP synergy.
